Effluent and receiving water samples which are going to be used in bioassays must be placed on ice or refrigerated immediately, and held at 4 degrees C until warmed for use.  

Tests must be initiated within 36 hours of collection of effluent samples.

If the sample contains organisms that may attack or be confused with the test organisms, the sample should be filtered through a 60 um mesh sieve before use.

Water quality parameters which should be measured in the sample before beginning a test include conductivity, pH, total residual chlorine, dissolved oxygen, alkalinity and hardness.

If the pH is outside the 6.0 - 9.0 range, the pH should be adjusted to 7.0 by careful addition of HCl or NaOH prior to use.
